My partner made a huge mistake and took 2mg for her very first dose last night around 9pm, instead of the 0.25mg she was supposed to start with.

By 1am she was already feeling nauseous, and by 3am she was violently vomiting. She's basically felt like absolute garbage all day today. We're pretty sure it's the Semaglutide, though there's a tiny chance it was a reheated dinner we had at 7pm, but the timing for a massive dose reaction seems more likely.

I'm assuming things will start to settle down over the next 72 hours as the levels in her system start to drop. From what I've read, 2mg is toward the high end of the maintenance range so it shouldn't be a "toxic" overdose, but obviously you're supposed to spend months building up to that.

Has anyone else ever done this or seen someone go through it? Any advice on how to manage the symptoms while we wait for it to wear off?

scary mistake but she should be okay - just a very unpleasant ride while it works through her system. definitely double check doses next time

oof that sounds rough my buddy did almost the same thing and was glued to the bathroom for like 2 days straight. small sips of water and electrolytes were the only thing he could keep down, plus just sleeping it off. it did pass but man he never skipped the starter dose again after that

Oof yeah jumping straight to 2mg is like skipping all the “training wheels” and going full send my friend accidentally doubled her dose once and was hugging the toilet for like 2 days. Small sips of fluids, electrolytes, and just letting her sleep it off helped the most. It should ease up but man that first hit can be brutal.

Oof that sounds rough. I’ve seen someone accidentally do a way-too-high first dose and it was basically a full day of couch+bucket mode. Best thing you can do is just keep her hydrated, small sips of water or electrolyte drinks, and bland foods if she can stomach anything. Anti-nausea stuff like ginger or dimenhydrinate helped my buddy a bit too. Honestly, it just takes time for it to taper off—72 hours sounds about right. Definitely start slow next time though, those first doses are no joke.

The "toxic" part isn't the drug itself, but the dehydration from the vomiting. Since she’s 24 hours in, she’s probably hit the peak, but she’s likely going to feel like a zombie for the next 4 to 5 days because the half-life is so long. 2mg is a massive shock to a "virgin" system.
