Math check for Semaglutide reconstitution

So, I’ve got these 2mg vials of semaglutide. I added 1ml of bac water to one and have been drawing to the 12.5 mark to hit that 250mcg dose. The thing is, the math says I should be getting 8 shots out of the vial, but I only managed to get 5 before it was empty.

I'm trying to figure out where I went wrong. Did I mess up the water ratio, or am I just misreading the syringe? It's frustrating because I'm burning through these way faster than I should be. Has anyone else had this happen, or am I just failing at basic peptide math?

sounds like syringe reading or dead space loss more than math being wrong. Those little bits left in the tip add up fast over multiple draws.

You’re probably losing some in the dead space of the syringe + what sticks to the vial walls. It adds up fast, especially with small vials.

Also depends what syringe you’re using. Some insulin pins waste way more than others, so the “on paper” doses never match real life.

I had the same thing happen my first time and thought I screwed up the math too turns out I was pulling a tiny air bubble each time without realizing.

If you injected a little air back in to equalize pressure each time, that can push liquid into places you can’t recover too. Not a huge amount but noticeable.

yeah I’ve totally done this before easiest mistake is just misreading the units on the insulin syringe… I ended up drawing too much a few times and wondered why the vial vanished so fast. double check the math and maybe mark the syringe with a sharpie for your dose, makes life way easier

The math actually checks out on paper, so you aren't crazy, but "syringe dead space" is probably eating your gains. Every time you pull a shot, a tiny bit of liquid stays trapped in the hub and the needle tip after you push the plunger. It doesn't look like much, but over 5 or 6 shots, that "waste" can easily add up to a quarter of a milliliter.

Yup, I’ve definitely done the same thing a few times. It’s usually either the syringe markings or just a tiny bit of leftover powder sticking to the vial that throws off the math. I’d double-check the actual units you’re drawing each time and maybe let the vial sit a minute so everything fully dissolves before measuring—makes a bigger difference than you’d think.

Across 8 planned shots, that "lost" liquid adds up fast, which is why your vial tapped out at 5. It’s super annoying because these peptides aren't exactly cheap. Another thing is "vial vacuum"—if you aren't injecting the same amount of air into the vial as the liquid you're pulling out, it gets harder and harder to get that last bit of moisture out of the corners.
